Description

RealHelix™ qPCR Kit is designed to perform the rapid real-time quantification of target from DNA or 1st-strand cDNA synthesized from RNA molecule. This kit includes a double strand DNA specific-binding fluorescent dye showing several-fold higher fluorescence than SYBR Green and little inhibition to the PCR process. The kit consists of convenient 2x concentrated premix solution and ROX passive dye. The 2x premix contains the hot-start PCR enzyme, dNTPs, buffers, Mg2+ and stabilizing agent. The hot-start PCR enzyme provides the high specific amplification of target DNA and minimizes the side products such as primer dimers. Included ROX dye could be used as a passive reference dye.

Quality Control

RealHelix™ qPCR Kit was evaluated by real-time PCR using the 10-fold serial-diluted human genomic DNA and a set of human gene-specific primer with ABI 7900HT Fast-real-time PCR system (Applied Biosystems, USA) and Rotor-Gene Q instrument (QIAGEN, Germany).

Figure 1. Comparison of real-time PCR using RealHelix™ qPCR Kit [Intercalator type] and Maxima SYBR qPCR Master Mix (Company F). The reactions were anayzed using human genomic DNAs (100, 10, 1 ng) isolated from whole blood. The accuracy of real-time PCR is demonstrated by R2 value = 0.9973 (RealHelix™ qPCR Kit) and R2 value = 0.9964 (Maxima SYBR qPCR Master Mix).

Comparison of real-time PCR using ReaHelix™ qPCR Kit [Probe type] and TaqMan Fast Universal PCR Master Mix (Company I).Human genomic DNAs (100, 10, 1 ng) isolated from whole blood were analyzed using beta-globin DNA-specific primer set and 5’-FAM-labelled probe. The accuracy of real-time PCR is denomstrated by R2 value = 0.99634 (RealHelix™ qPCR Kit) and R2 value= 0.99917 (TaqMan Fast Universal PCR Master Mix).

Absolute quantification by RealHelix™ qPCR Kit. 10-fold serial-diluted human genomic DNA as the standard sample and 25 ng human genomic DNA as test sample were analyzed using the RealHelix™ qPCR Kit for estrogen receptor gene-specific primer set. The accuracy of real-time PCR using RealHelix™ qPCR Kit is demonstrated by R2 value = 0.99649.
